Profil Fitokimia dan Aktivitas Antibakteri Ulva Lactuca Hasil Ekstraksi Bertingkat Berbasis Ultrasound-Assisted Extraction

Abstract


Produksi rumput laut di Indonesia cukup melimpah, namun pemanfaatannya terutama Ulva lactuca masih belum optimal, khususnya sebagai sumber senyawa bioaktif. Penelitian ini bertujuan untuk mengidentifikasi kandungan metabolit sekunder serta mengevaluasi aktivitas antibakteri ekstrak Ulva lactuca yang diperoleh melalui ekstraksi bertingkat. Metode penelitian meliputi preparasi sampel, ekstraksi bertingkat menggunakan pelarut n-heksana, etil asetat, dan etanol dengan bantuan teknik Ultrasound-Assisted Extraction (UAE), skrining fitokimia secara kualitatif, serta uji aktivitas antibakteri menggunakan metode difusi agar terhadap bakteri Staphylococcus aureus dan Escherichia coli. Hasil penelitian menunjukkan bahwa ekstrak Ulva lactuca mengandung senyawa alkaloid, flavonoid, terpenoid dan fenol. Ketiga ekstrak Ulva lactuca menunjukkan kemampuan dalam menghambat pertumbuhan bakteri. Adanya efek sinergis antar senyawa bioaktif dalam Ulva lactuca mampu membentuk kompleks dengan protein ekstraseluler dan terlarut sehingga merusak membran sel bakteri. Secara keseluruhan, kombinasi ekstraksi bertingkat dan teknik UAE terbukti efektif dalam mengisolasi senyawa bioaktif dari Ulva lactuca dan meningkatkan potensi pemanfaatannya sebagai sumber antibakteri alami.
